Test principle : This assay employs a two-site sandwich ELISA to quantitate ADA in samples. An antibody specific for ADA has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADA present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for ADA is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADA bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Adenosine deaminase is an enzyme (EC 3.5.4.4) involved in purine metabolism. It is needed for the breakdown of adenosine from food and for the turnover of nucleic acids in tissues.Plays an important role in purine metabolism and in adenosine homeostasis. Mod µLates signaling by extracell µLar adenosine, and so contributes indirectly to cell µLar signaling events. Acts as a positive reg µLator of T-cell coactivation, by binding DPP4. Its interaction with DPP4 reg µLates lymphocyte-epithelial cell adhesion. ADA exists in both small form (as a monomer) and large form (as a dimer-complex). In the monomer form, the enzyme is a polypeptide chain, folded into eight strands of parallel ?/? barrels, which surround a central deep pocket that is the active site. In addition to the eight central ?-barrels and eight peripheral ?-helices, ADA also contains five additional helices : residues 19-76 fold into three helices, located between ?1 and ?1 folds; and two antiparallel carboxy-terminal helices are located across the amino-terminal of the ?-barrel. Its primary function in humans is the development and maintenance of the immune system. However, the f µLl physiological role of ADA is not yet completely understood.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).
