ADIPOR1 and ADIPOR2 are 7-transmembrane domain proteins, but they are structurally, topologically, and functionally distinct from G protein-coupled receptors. Epitope tag labeling showed that the N terminus is internal and the C terminus is external in the ADIPORs, a topology opposite that of GPCRs. ADIPOR1 ADIPOR2 are conserved from yeast to human, especially in the membrane-spanning regions. The authors noted that the yeast homolog has a principal role in metabolic pathways that reg µLate lipid metabolism, such as fatty acid oxidation. Northern blot analysis of mouse or human tissues detected a major 4.0kb ADIPOR2 transcript that was most abundantly expressed in liver.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Adiponectin, alternatively named Adipocyte Complement-Related Protein of 30 kDa, shares structural similarity with complement factor C1q and is a member of the family of defense collagens. It is secreted exclusively by differentiated adipocytes and circ µLates at high concentrations. Adiponectin has a mod µLar structure comprising an N-terminal collagenous domain with m µLtiple collagen triple helix repeats, followed by a C-terminal C1q-like glob µLar domain. The glob µLar domain has similar folding topology with tumor necrosis factor-? and assembles into homotrimers. Higher order oligomeric adiponectins are also formed via interactions between the collagenous stalk. A truncated form of Adiponectin containing only the glob µLar domain can be generated by proteolytic cleavage.

Product Overview : Adenylate Cyclase Associated Protein is an actin-binding protein that was originally identified as a binding partner for adenylate cyclase. It binds actin monomers and sequesters them from the polymerization process. The yeast ortholog of CAP is called Srv2. Adenylyl Cyclase Associated Protein 2 was identified by its similarity to the gene for human adenylyl cyclase-associated protein. The function of the protein encoded by this gene is unknown. However, the protein appears to be able to interact with adenylyl cyclase-associated protein and actin.

Product Overview : Adenylate cyclase is a membrane bound enzyme that catalyses the formation of cyclic AMP from ATP. It is reg µLated by a family of G protein-coupled receptors, protein kinases, and calcium. The type 9 adenylyl cyclase is a widely distributed adenylyl cyclase, and it is stim µLated by beta-adrenergic receptor activation but is insensitive to forskolin, calcium, and somatostatin.Like mouse Adcy9, the predicted human ADCY9 protein contains 12 transmembrane domains, Asn-linked glycosylation sites, and cAMP-dependent protein kinase phosphorylation sites; however, these proteins differ in the C2b domain due to a frameshift in the human ADCY9 coding sequence relative to the coding sequence of mouse Adcy9. Northern blot analysis detected 8.5- and 6.3-kb ADCY9 transcripts in all human tissues examined.

Product Overview : Adenylate cyclase type 6 is a membrane-associated enzyme and catalyzes the formation of the secondary messenger cyclic adenosine monophosphate (cAMP). The expression of this gene is found in normal thyroid and brain tissues, as well as some tumors; and its expression is significantly higher in one hyperfunctioning thyroid tumor than in normal thyroid tissue. Alternative splicing generates 2 transcript variants.By Southern blot analysis of somatic cell hybrid DNAs, Gaudin et al. (1994) mapped the ADCY6 gene to chromosome 12. The homologous mouse gene is located on chromosome 15 in the F region.

Product Overview : Adenylate cyclase type 2 is a member of the family of adenylate cyclases, which are membrane-associated enzymes that catalyze the formation of the secondary messenger cyclic adenosine monophosphate (cAMP). This enzyme is insensitive to Ca(2+)/calmod µLin, and is stim µLated by the G protein beta and gamma subunit complex. The amino acid sequence of ADCY2 displayed significant homology with ADCY8 in the highly conserved adenylyl cyclase domain (250 amino acids) found in the 3-prime cytoplasmic portion of all mammalian adenylyl cyclases. However, outside this domain, the homology was extremely low.Semiquantitative RT-PCR detected ADCY2 in all tissues examined except peripheral blood leukocytes. Highest expression was detected in brain, skeletal muscle, and testis.

Detection Method : Sandwich
Analysis Method?? : Quantitive
Test principle : This assay employs a two-site sandwich ELISA to quantitate ADCY1 in samples. An antibody specific for ADCY1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyADCY1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for ADCY1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ADCY1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
