Background : Catalytic subunit of a constitutively active serine/threonine-protein kinase complex that phosphorylates a large number of substrates containing acidic residues C-terminal to the phosphorylated serine or threonine. Reg µLates numerous cell µLar processes, such as cell cycle progression, apoptosis and transcription, as well as viral infection. May act as a reg µLatory node which integrates and coordinates numerous signals leading to an appropriate cell µLar response. During mitosis, functions as a component of the p53/TP53-dependent spindle assembly checkpoint (SAC) that maintains cyclin-B-CDK1 activity and G2 arrest in response to spindle damage. Also required for p53/TP53-mediated apoptosis, phosphorylating \\\'Ser-392\\\' of p53/TP53 following UV irradiation. Can also negatively reg µLate apoptosis. Phosphorylates the caspases CASP9 and CASP2 and the apoptotic reg µLator NOL3. Phosphorylation protects CASP9 from cleavage and activation by CASP8, and inhibits the dimerization of CASP2 and activation of CASP8. Reg µLates transcription by direct phosphorylation of RNA polymerases I, II, III and IV. Also phosphorylates and reg µLates numerous transcription factors including NF-kappa-B, STAT1 CREB1 IRF1 IRF2, ATF1 SRF, MAX, JUN, FOS, MYC and MYB. Phosphorylates PML at \\\'Ser-565\\\' and primes it for ubiquitin-mediated degradation. Plays an important role in the circadian clock function by phosphorylating ARNTL/BMAL1 at \\\'Ser-90\\\' which is pivotal for its interaction with CLOCK and which controls CLOCK nuclear entry.

Background : Non-receptor tyrosine-protein kinase that plays an important role in the reg µLation of cell growth, differentiation, migration and immune response. Phosphorylates tyrosine residues located in the C-terminal tails of Src-family kinases (SFKs) including LCK, SRC, HCK, FYN, LYN or YES1. Upon tail phosphorylation, Src-family members engage in intramolec µLar interactions between the phosphotyrosine tail and the SH2 domain that res µLt in an inactive conformation. Background : Tyrosine-protein kinase that acts as cell-surface receptor for CSF1 and IL34 and plays an essential role in the reg µLation of survival, proliferation and differentiation of hematopoietic precursor cells, especially mononuclear phagocytes, such as macrophages and monocytes. Promotes the release of proinflammatory chemokines in response to IL34 and CSF1 and thereby plays an important role in innate immunity and in inflammatory processes. Plays an important role in the reg µLation of osteoclast proliferation and differentiation, the reg µLation of bone resorption, and is required for normal bone and tooth development. Required for normal male and female fertility, and for normal development of milk ducts and acinar structures in the mammary gland during pregnancy. Promotes reorganization of the actin cytoskeleton, reg µLates formation of membrane ruffles, cell adhesion and cell migration, and promotes cancer cell invasion. Activates several signaling pathways in response to ligand binding. Phosphorylates PIK3R1 PLCG2, GRB2, SLA2 and CBL.
