Background : nnate immune receptor which acts as a cytoplasmic sensor of viral nucleic acids and plays a major role in sensing viral infection and in the activation of a cascade of antiviral responses including the induction of type I interferons and proinflammatory cytokines. Its ligands include : 5\'-triphosphorylated ssRNA and dsRNA and short dsRNA (<1 kb in length). In addition to the 5\'-triphosphate moiety, blunt-end base pairing at the 5\'-end of the RNA is very essential. Overhangs at the non-triphosphorylated end of the dsRNA RNA have no major impact on its activity. A 3\'overhang at the 5\'triphosphate end decreases and any 5\'overhang at the 5\' triphosphate end abolishes its activity. Upon ligand binding it associates with mitochondria antiviral signaling protein (MAVS/IPS1) which activates the IKK-related kinases : TBK1 and IKBKE which phosphorylate interferon reg µLatory factors : IRF3 and IRF7 which in turn activate transcription of antiviral immunological genes, including interferons (IFNs); IFN-alpha and IFN-beta. Detects both positive and negative strand RNA viruses including members of the families Paramyxoviridae : Human respiratory syncytial virus and measles virus (MeV), Rhabdoviridae : vesic µLar stomatitis virus (VSV), Orthomyxoviridae : influenza A and B virus, Flaviviridae : Japanese encephalitis virus (JEV), hepatitis C virus (HCV), dengue virus (DENV) and west Nile virus (WNV). It also detects rotavirus and reovirus. Also involved in antiviral signaling in response to viruses containing a dsDNA genome such as Epstein-Barr virus (EBV). Detects dsRNA produced from non-self dsDNA by RNA polymerase III, such as Epstein-Barr virus-encoded RNAs (EBERs). May play important roles in gran µLocyte production and differentiation, bacterial phagocytosis and in the reg µLation of cell migration.

Background : ATP-dependent RNA helicase. Binds to partially double-stranded RNAs (dsRNAs) in order to unwind RNA secondary structures. Unwinding is promoted in the presence of single-strand binding proteins. Mediates also RNA duplex formation thereby displacing the single-strand RNA binding protein. Background : Probable ATP-dependent RNA helicase required during spermatogenesis (PubMed : 10920202, PubMed : 21034600). Required to repress transposable elements and preventing their mobilization, which is essential for the germline integrity. Acts via the piRNA metabolic process, which mediates the repression of transposable elements during meiosis by forming complexes composed of piRNAs and Piwi proteins and governs the methylation and subsequent repression of transposons. Involved in the secondary piRNAs metabolic process, the production of piRNAs in fetal male germ cells thro µgh a ping-pong amplification cycle (By similarity).

Background : M µLtifunctional ATP-dependent RNA helicase. The ATPase activity can be stim µLated by various ribo- and deoxynucleic acids indicative for a relaxed substrate specificity. In vitro can unwind partially double-stranded DNA with a preference for 5\'-single-stranded DNA overhangs. Is involved in several steps of gene expression, such as transcription, mRNA maturation, mRNA export and translation. However, the exact mechanisms are not known and some functions may be specific for a subset of mRNAs. Involved in transcriptional reg µLation. Can enhance transcription from the CDKN1A/WAF1 promoter in a SP1-dependent manner. Found associated with the E-cadherin promoter and can down-reg µLate transcription from the promoter. Involved in reg µLation of translation initiation. Proposed to be involved in positive reg µLation of translation such as of cyclin E1/CCNE1 mRNA and specifically of mRNAs containing complex secondary structures in their 5\'UTRs; these functions seem to require RNA helicase activity.
