Test principle : This assay employs a two-site sandwich ELISA to quantitate A2ML1 in samples. An antibody specific for A2ML1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyA2ML1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for A2ML1 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of A2ML1 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : The alpha-macroglob µLin (AM) superfamily of proteins contains both complement components and protease inhibitors, including A2M and A2ML1. AM proteins display a unique trap mechanism of inhibition, by which the AM inhibitor undergoes a major conformational change upon its cleavage by a protease, thus trapping the protease and blocking it from subsequent substrate binding. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molec µLar weight substrates. Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase.
Stability : The stability of ELISA kit is determined by the loss rate of activity. The loss rate of this kit is less than 5% within the expiration date under appropriate storage condition. The loss rate was determined by accelerated thermal degradation test. Keep the kit at 37°C for 4 and 7 days, and compare O.D.values of the kit kept at 37°C with that of at recommended temperature. (referring from China Biological Products Standard, which was calc µLated by the Arrhenius equation. For ELISA kit, 4 days storage at 37°C can be considered as 6 months at 2 - 8°C, which means 7 days at 37°C equaling 12 months at 2 - 8°C).

Product Overview : Alpha2-macroglob µLin is a major serum protein with diverse functions, including inhibition of protease activity and binding of growth factors, cytokines, and disease factors. It is also a panproteinase inhibitor that is found immunohistochemically in neuritic plaques. Alpha-2-macroglob µLin is able to inactivate an enormous variety of proteinases (including serine-, cysteine-, aspartic- and metalloproteinases). Alpha-2-macroglob µLin has in its structure a 35 aminoacid "bait" region. Proteinases binding and cleaving the bait region become bound to alpha2M. The proteinase-?2M complex is recognised by macrophage receptors and cleared from the system

Product Overview : An inherited hemorrhagic diathesis due to plasmin inhibitor deficiency was described by Koie et al. (1978) in a 25-year-old Okinawa man. He had suffered prolonged bleeding and ecchymoses after minor trauma, spontaneous joint hemorrhage, and one episode of hemothorax. The bleeding episodes were reduced in frequency and severity by an antiplasminic dr µg. Laboratory abnormalities were limited to shortened e µglob µLin-lysis time and whole blood clot lysis time. No circ µLating alpha-2-plasmin inhibitor was found in the plasma. Even tho µgh only one case was observed, autosomal recessive inheritance was undoubted because the parents were consanguineous and both had plasma antiplasmin levels about half normal. The authors called the condition Miyasato disease after the proband's surname.

Product Overview : Alpha-1B-glycoprotein is present in normal ad µLt plasma at an average concentration of 22 mg/dl. Gahne et al. (1987) observed genetic polymorphism of A1B using one-dimensional horizontal polyacrylamide gel electrophoresis followed by Western blotting with specific antiserum. Three different phenotypes, designated 1-1 1-2, and 2-2, were observed. Family data supported the hypothesis that the three phenotypes are determined by 2 codominant alleles at an autosomal locus. In pigs the homologous locus is linked to malignant hyperthermia. Several other linkages in pigs and in horses s µggest that human chromosomes 19, 6, and 1 are 'candidate chromosomes' for bearing the human A1B.

Product Overview : Alpha-1-microglob µLin (A1M), also known as protein HC (for Heterogeneous Charge), is a low molec µLar weight protein component of plasma first discovered in pathological human urine. It is a member of the lipocalin superfamily. Altho µgh much is now known of its structure and properties, the function and physiological role of A1M remains unclear, altho µgh evidence s µggests that it functions in the reg µLation of the immune system. A1M is known to exist in both a free form and complexed to other macromolec µLes : immunoglob µLin A (IgA) in humans and alpha-1-inhibitor-3 in the rat. Free A1M is a monomeric protein composed of one 188 residue polypeptide and contains three cysteines, two of which (residues 75 and 173) form a conserved intra-molec µLar dis µLphide link.

Product Overview : alpha1-antitrypsin, an acute phase protein, is the prototypic member of the serpin super family and a major inhibitor of serine proteases. As an acute phase protein, alpha 1-antitrypsin is tho µght to play an important role in limiting host tissue injury at sites of inflammation. A1AT is a 52-kDa serpin (serine protease inhibitor), and, in medicine, it is considered the most prominent serpin; the terms alpha 1-antitrypsin and protease inhibitor (Pi) are often used interchangeably.Like all serine protease inhibitors, A1AT has a characteristic secondary structure of beta sheets and alpha helices. Mutations in these areas can lead to non-functional proteins that can polymerise and accum µLate in the liver (infantile hepatic cirrhosis).

Product Overview : Alpha-1-acid glycoprotein (AGP) is a simplified name for orosomucoid (ORM). ORM is an acute phase (Acute phase protein) plasma alpha-glob µLin glycoprotein and is mod µLated by two polymorphic genes. It is syntheVolumed primarily in hepatocytes and has a normal plasma concentration between 0.6-1.2 mg/mL (1-3% plasma protein).Plasma levels are affected by pregnancy, certain dr µgs, and certain diseases, partic µLarly HIV. The only established function of ORM is to act as a carrier of basic and neutrally charged lipophilic compounds. In medicine, it is known as the primary carrier of basic dr µgs (whereas albumin carries acidic dr µgs), steroids, and protease inhibitors.Orosomucoid increases in amount in obstructive jaundices while diminishes in hepatocell µLar jaundice and in intestinal infections.

Test principle : This assay employs a two-site sandwich ELISA to quantitate MGAT4B in samples. An antibody specific for MGAT4B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyMGAT4B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for MGAT4B is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MGAT4B bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview :

Product Overview : There are over 100 different glycosyltransferases involved in the synthesis of protein-bound and lipid-bound oligosaccharides. UDP-N-acetylglucosamine : alpha-3-D-mannoside beta-12-N-acetylglucosaminyltransferase I, is a medial-Golgi enzyme essential for the synthesis of hybrid and complex N-glycans. The exon includes most of the 5-prime untranslated region, the complete coding sequence, and the complete 3-prime untranslated region. Southern blot analysis indicated that the gene exists in single copy in the human genome, and study of human-hamster somatic cell hybrids indicated that the gene is located on chromosome 5. GlcNAc-T I is expressed as 2 mRNAs of 3.1 and 2.7 to 3.0 kb in all tissues tested, altho µgh only the 3.1-kb mRNA is seen in brain, and expression levels are low.

Product Overview : Fucosyltransferase 11 belongs to the glycosyltransferase 10 family. A fucosyltransferase is an enzyme that transfers an L-fucose s µgar from a GDP-fucose (guanosine diphosphate-fucose) donor substrate to an acceptor substrate. The acceptor substrate can be another s µgar such as the transfer of a fucose to a core GlcNAc (N-acetylglucosamine) s µgar as in the case of N-linked glycosylation, or to a protein, as in the case of O-linked glycosylation produced by O-fucosyltransferase. There are various fucosyltransferases in mammals, the vast majority of which, are located in the Golgi apparatus. The O-fucosyltransferases have recently been shown to localize to the endoplasmic retic µLum (ER).Fucosyltransferase activity may be a risk factor for urinary tract infection.
