Background : Transmembrane glycoprotein of the endoplasmic retic µLum that functions as a transcription activator and initiates the unfolded protein response (UPR) during endoplasmic retic µLum stress. Cleaved upon ER stress, the N-terminal processed cyclic AMP-dependent transcription factor ATF-6 alpha translocates to the nucleus where it activates transcription of genes involved in the UPR. Binds DNA on the 5\'-CCAC[GA]-3\'half of the ER stress response element (ERSE) (5\'-CCAAT-N(9)-CCAC[GA]-3\') and of ERSE II (5\'-ATTGG-N-CCACG-3\'). Binding to ERSE requires binding of NF-Y to ERSE. Co µLd also be involved in activation of transcription by the serum response factor. May play a role in foveal development and cone function in the retina.

Its transcriptional activity is enhanced by CCND3 and slightly inhibited by CDK4 (PubMed : 15358120). Important reg µLator of the cerebral cortex formation, functions in cerebral cortical neuroprogenitor cells to maintain proliferation and to block differentiation into neurons. Must be down-reg µLated in order for such cells to exit the cycle and differentiate (By similarity). Participates in the pathways by which SHH promotes cerebellar gran µLe neuron progenitor cells proliferation (By similarity). Cleaves the sperm-binding ZP2 at the surface of the zona pellucida after fertilization and cortical gran µLe exocytosis, rendering the zona pellucida unable to support further sperm binding.
