Test principle : This assay employs a two-site sandwich ELISA to quantitate ACTA2 in samples. An antibody specific for ACTA2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and anyACTA2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conj µgated antibody specific for ACTA2 is added to the wells. After washing, Streptavidin conj µgated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ACTA2 bound in the initial step. The color development is stopped and the intensity of the color is measured.
Product Overview : Alpha-actin-2 also known as actin, aortic smooth muscle or alpha smooth muscle actin (?-SMA, SMactin, alpha-SM-actin, ASMA) is a protein that in humans is encoded by the ACTA2 gene located on 10q22-q24. Actin alpha 2, the human aortic smooth muscle actin gene, is one of six different actin isoforms which have been identified. Actins are highly conserved proteins that are involved in cell motility, structure and integrity. Alpha actins are a major constituent of the contractile apparatus. Alpha-smooth muscle actin (?-SMA) is commonly used as a marker of myofibroblast formation (Nagamoto et al., 2000).

Product Overview : Beta-actin (gene name ACTB) is one of the two nonmuscle cytoskeletal actins. Actins are highly conserved proteins that are involved in cell motility, structure and integrity. Alpha actins are a major constituent of the contractile apparatus. In a neoplastic cell line res µLting from treatment of c µLtured human diploid fibroblasts with a chemical mutagen, Leavitt et al. (1982) observed a mutant form of beta actin. Toyama and Toyama (1984) isolated and characterized lines of KB cells resistant to cytochalasin B. They found that one resistant line had an alteration in beta-actin. Such cells bound less cytochalasin B than did parental KB cells. The authors s µggested that the primary site of action of cytochalasin B on cell motility processes is beta-actin.

Product Overview : The isoprostanes are a family of eicosanoids of non-enzymatic origin produced by the random oxidation of tissue phospholipids by oxygen radicals. Isoprostanes appear as artifacts in tissue and plasma samples which have undergone oxidative degradation during prolonged or improper storage. They also appear in the plasma and urine under normal conditions and are elevated by oxidative stress. At least one of the isoprostanes, 8-isoprostane (8-epi PGF2α), has been shown to have biological activity. It is a potent p µLmonary and renal vasoconstrictor1 and has been implicated as a causative mediator of hepatorenal syndrome and p µLmonary oxygen toxicity.2 8-Isoprostane has been proposed as a marker of antioxidant deficiency and oxidative stress and elevated levels have been found in heavy smokers.3 8-Isoprostane levels are also a relative indicator of sample integrity for lipid-containing samples such as serum, plasma, and whole cell preparations.
