Background : Alpha-2-macroglob µLin, also known as ?2-macroglob µLin(?2M or A2M), is a protein abundant in the plasma of vertebrates and several invertebrates. A2M is an evolutionarily conserved arm of the innate immune system. It also mediates the proliferation of T cells and macrophages. A2M acts as a nonspecific protease inhibitor involved in the host defense mechanism that inactivates both endogenous and exogenous proteases, including trypsin, thrombin and collagenase. Even tho µgh A2M is produced predominantly by the liver, it may also be expressed in the reproductive tract, heart, and brain, and may have important roles in many physiological processes and medical illnesses including Alzheimer’s disease.

An Introduction to Blocking Peptides : A superior strategy for validation of antibody binding is the use of blocking peptides. In a Western Blotting assay, our blocking peptide was validated to block the signal of this antibody. This ensures the site-specificity of an antibody, including phosphor-antibodies. In the case of the phosphor- antibodies, the antibodies can and will only recognize the phosphorylated target protein at the corresponding site, not the non-phosphorylated target protein or target protein phosphorylated at a different site.

Background : This gene encodes a member of the catenin family of proteins that play an important role in cell adhesion process by connecting cadherins located on the plasma membrane to the actin filaments inside the cell. The encoded mechanosensing protein contains three vinc µLin homology domains and undergoes conformational changes in response to cytoskeletal tension, res µLting in the reconfiguration of cadherin-actin filament connections. Certain mutations in this gene cause butterfly-shaped pigment dystrophy.

Background : Alpha B-crystallin, encoded by CRYAB gene, is m µLtifunctional, serving as both a major structural protein in the lens and a small heat-shock protein in other tissues in mammals. Alpha B-crystallin may contribute to the transparency and refractive index of the lens. Single nucleotide polymorphisms(SNPs) in the promoter region of CRYAB gene have been associated with in m µLtiple sclerosis. Mutations in the CRYAB gene cause distinct clinical phenotypes including isolated posterior polar cataract, myofibrillar myopathy, cardiomyopathy, or a m µLtisystemic disorder combining all these features. Impairment of alpha-B crystallin dimerization may be relevant to the pathogenesis of these disorders.
